I have given more thought to our GenGen program schedule and have
suggestion/requests.

I expect Bob's discussion to be primarily on the various databases of
Ancestry.com and very little on the DNA features.  That is one reason I
suggested you step in the breach and do something on DNA at Ancestry.

In addition being a transition, I suggested that your discussion could
permit time for followon questions from your December presentation.

Rather than a lecture in November,I conducted more of a workshop Y-DNA
and did everything live on the Internet. It was purposefully very
elementary for some of our members who have been askingfor such a
presentation. Although I had a plan available but did not use any
handouts, but rather just went along with questions.  I just logged on
to my personal account and looked at the various features while
explaining the details.  For example, I explained haplotype and then
went into what an STR was and associated complications. I sent into
haplotree explaining what a SNP was.  I stayed on each feature until
either the questions ran out or a question lead us to another feature.
I got several expressions of appreciation, especially from those who had
been asking for beginner sessions. I have ask Bob to do a similar
presentation of using the databases of Ancestry.com

In December you presented a well received presentation of using the
results of autosomal tests, but did not discuss the interfaces to the
autosomal data itself.  I have made such presentations in the past, but
not as a workshop. Perhaps, you could do a workshop on accessing the
autosomal data.  We don't need as muc background as you presented in
December, but start by simply logging into an account and start
accessing data, explaining what you find.  As I told Bob, I would be
glad to expose my account and would even do the keyboarding you you can
just talkand tell me what feature to go to next.  I now have autosomal
data at 12aandMe, FTDNA, and Ancestry, but Iam still learning.  (For
years, I have concentrated in developing phylogenies and doing other
basic research for mtDNA Haplogroup J and Tand have neglected my own
geneaology. I just getting back to it.)

What to you think?

I have just put the near finishing touches on a paper entitled:

A Comparative Analysis of the RCC Correlation and FamilyTreeDNA TiPTM
Probability Approaches for Estimating the Time to the Most Recent Common
Ancestor of a Pair of Y-DNA Haplotypes.

I have sent it to Bennett Greenspan suggesting to him that if he were to show it
to his staff and his expert consultants, and if they would have any comments on
the analysis, I would be pleased to consider incorporating them into changes in
the paper, or to add some appropriate comments in endnotes, etc.

I very much appreciate the discussions I have had with some members of this
GenGen group, and particularly with Sidney Sachs and Elizabeth Waltman as the
analysis progressed. Nell Hurley kindly sent me information on her Pettygrews
and Linda McKee sent me information on her Kee and McKees.

I would be very pleased if any member of this GenGen forum will send me their
own comments and I will treat them in the same way as whatever FTDNA sends me.

I was curious if anyone can comment on some y-DNA results from my DNA project

I recently received y-DNA results back from my 5th cousin.  According to our
respective family trees, we trace back to the same 4th great-grandfather.  Below
are differences we have:

For the first 12 markers we match 12 for 12.
At the 25 marker level everything matches except for

DYS459
my Kit 40543: 9-10
my cousin's Kit 269385: 9-9

DYS464
my Kit 40543: 11-12-14-15-16
my cousin's Kit 269385: 11-12-12-14-14-15-15-16

At the 37 marker level there is a difference at

DYS-CDY
my Kit 40543: 35-41
my cousin's Kit 269385: 34-35

The difference at DYS459 is not extraordinary.

However, I don't know how I should interpret the differences at DYS464 and
DYS-CDY.  If the paper trail that my cousin and I have is correct, would such
diverse differences occur?

When the next sale occurs, I think I should upgrade to the 67 marker or the 111
marker level.

Tom,

 

First the DYS#464 results.  I suspect that your 464 results are not correct.  DYS #464 is a very difficult one to read.  So, I suspect what happened is that they had some trouble reading yours and misjudged the peaks.

 

CDYa/b is highly unusual.  This is a marker with a very high mutation rate, so differences between relatives is not unusual.  However, the amount of difference in your case is unusual.

 

I would contact FTDNA and ask them to reexamine 464 and CDY for you and your cousin.  Is there is still a difference at 464, I would do a 646x test on both of you.  This will read the correct number of duplicates.

Looking at FTDNA's website, I believe I should use the infinite allele
comparison method to interpret the results.

Thus, DYS459 would be considered a mismatch and would be interpreted as a
Genetic Distance of 1.  Is this correct?

The DYS464 differences would be considered a palindromic copy number change
where one kit is DYS464abcde and the other kit is DYS464abcdefgh.  This would be
interpreted as a Genetic Distance of 3.  Is this correct?

DYS-CDY would be also be considered a mismatch and would be interpreted as a
Genetic Distance of 1.  Is this correct?

Thus, the total Genetic Distance between the two kits would be 5.  Is this
correct?

Does this indicate that my cousin and I are descended from the same male
ancestor?

For 5th cousins, about 20% of the times, one would get with 3 markers off.  What I found with your value on 464 is that you have an odd number of copies.  I think  I never saw that before. 
 
About updating to more markers, I do not think that is what I would do.  I think checking your cousin on the SNP M458 that defined you as R1a1a1g may be the better way of getting  your answer.
